How to choose PCR tubes?

In our last article, we talked about PCR and the factors that affect PCR amplification. Next, Peky Biotech is honored to share the selection of PCR tubes.


1. Material selection

Polypropylene (PP) is generally used as the material for both ordinary PCR and real-time PCR (qPCR). Due to its biological inertness, it is not easy to adhere to molecules on its surface and has good chemical tolerance and temperature tolerance. PCR consumables are usually in direct contact with reagents or samples, so high quality materials and good processing techniques are required in the production and preparation process.

2. Standard Profile or Low Profile 

The low profile consumables are lower in height. This design minimizes the space above the reaction system, improves the heat conduction efficiency and reduces the evaporation effect during the thermal cycle.

3. Flat Cap or Domed Cap

Flat Cap can provide accurate fluorescence signal transmission for qPCR. Meanwhile, it’s easy to write on it.

When contacting with PCR instrument cover, domed cap can prevent the tube from deformation caused by pressure. However, it will affect the fluorescence signal transmission and cannot be applied in qPCR experiment.

4. Clear tube or White tube

PCR tubes are often available in a variety of color formats to facilitate visual differentiation and identification of samples, especially in high-throughput experiments. Although the color of the tubes has no effect on DNA amplification, it is recommended to use white tubes rather than clear tubes for sensitive and accurate fluorescence detection when setting up real-time PCR or qPCR reactions. Cause the white PCR tubes can reflect the fluorescence signal to the maximum extent, reduce the cross-contamination of the signal between the wells, and optimize the results of real-time fluorescence quantitative PCR.

5. Cleanliness of PCR Tubes

PCR consumables must be free of contaminants and inhibitors. High pressure and radiation can remove bacteria and DNA enzymes, but not dust and DNA residues, which may inhibit PCR reactions or produce nonspecific amplification. Substances have absorbance, even small pollutants will absorb the fluorescence in the reaction system, affecting the collection of fluorescence signal. Therefore, in the production of these plastic consumables, from molding to final packaging must be carried out in a facility environment with particle number limits.
